Selection of the proper signal sequence for recombinant protein production in mammalian and insect cell culture is a crucial step that should not be overlooked during construct design.

If the protein of interest contains a native signal sequence, this can be retained in the expression construct design. Many signal sequences are functional in heterologous organisms, which is convenient for recombinant production. If recombinant expression and/or secretion is low with the native sequence, it can be replaced with a heterologous signal sequence. For example, a signal sequence that is native to the insect or mammalian expression host cell line may improve recombinant production of the protein of interest. It is wise to design multiple constructs with native and heterologous signal sequences to determine empirically which one works best.
