Fusion proteins are often used to increase the expression and solubility of the target protein. However, a relatively large fusion partner may negatively affect the functionality of the target protein in biochemical or structural studies. Inclusion of a protease-specific cleavage site in the expression construct can facilitate removal of the fusion partner during purification.
